The University of Uyo was established in 1991 by the Nigerian Federal Government. Prior to that, there was the University of Cross River State in Uyo, which was founded in 1981 by the Cross River State Government. In 1987, Akwa Ibom State was created out of Cross River State, with Uyo as its headquarters. From that point, until the Federal Government took over in 1991, the University was jointly owned by the Akwa Ibom and Cross River State Governments.

Initially proposed as a Federal University of Technology (FUT), the plan was for the Uyo campus of the University of Cross River State to become the nucleus of the proposed university. However, after discussions and implementation strategy meetings, it was decided in a letter dated September 24, 1991, that the University of Cross Rivers State should be converted into a Conventional University instead of a FUT. Academic activities began in the 1991/92 session.

The University of Uyo’s historical transformation is both humble and unique. The campus on which it is located started as a practicing school in the 1930s and became a Teacher Training College in the 1940s. It was later upgraded to an Advanced Teachers’ Training College in the 1960s.
